Human Antithrombin is derived from human plasma, purified through affinity chromatography and gel filtration chromatography methods, and subjected to S/D virus inactivation steps.

Human Antithrombin is a serine protease inhibitor that forms a complex with heparin, leading to the inactivation of factor IIa (thrombin) and factor Xa. The biochemical potency of heparin can be determined based on the catalytic dissociation rate of residual coagulation factors on the corresponding chromogenic substrates.

①Heparinases

Heparinases can clave glycosidic bonds of heparin and/or heparan sulfate by a β-elimination mechanism, generating unsaturated products (mostly 

disaccharides) with a double bond be-tween C4 and C5 of the uronate residue.

How to use

The lyophilized human antithrombin is stored at -20 °C and has an expiration date of more than 12 months;

1 IU/mL human antithrombin solution should be stored at 2-8°C protected from light and had an expiration date of more than 1 month.

About the Hair Color Substrate Method

Human antithrombin is commonly used to test the potency of heparin (chromogenic substrate method).

chromogenic substrates are chemically synthesized small peptides with a chromogenic group (pNA) at one end. Specific enzymes, such as thrombin, catalyze the separation of pNA. Free pNA can be detected by spectrophotometer or microplate reader to determine the activity of the corresponding substance.

Common chromogenic substrates are: thrombin pigment substrate S-2238, FXa chromogenic substrate S-2765 and S-2222.

Chromogenic assays are currently the main heparin titer detection method, which is the detection method of heparin (especially low molecular weight heparin) specified in the European Pharmacopoeia (EP) and the United States Pharmacopoeia (USP), and will be included in the 2015 edition of the Chinese Pharmacopoeia. This method is also widely used in laboratory studies and in vitro diagnostics.

By controlling the conditions of the reaction system, the chromogenic substrate method can detect coagulation factors, cofactors, or inhibitors of coagulation factors.
