The Asnail™ Heparin Anti-Factor 10 Assay is a two-step chromogenic substrate assay for the detection of heparin activity (including unfractionated heparin and low molecular weight heparin) in manual or automated experiments. This method is only suitable for the detection of heparin in human citric acid plasma or purified solution.

The kit is intended for scientific research only and is prohibited for clinical diagnosis and treatment.

①Heparinases

Heparinases can clave glycosidic bonds of heparin and/or heparan sulfate by a β-elimination mechanism, generating unsaturated products (mostly 

disaccharides) with a double bond be-tween C4 and C5 of the uronate residue.

Heparin is a sulfated polysaccharide that has a high affinity for thrombin. When combined with heparin, antithrombin exhibits rapid and effective inhibitory activity against coagulation factors such as serine proteases IXa, Xa, and thrombin. Low-molecular-weight heparin (LMWH) and heparin analogues, such as dalteparin sodium, inhibit factor Xa activity more effectively than thrombin activity, while unfractionated heparin (UFH) effectively inhibits thrombin and other serine protease activity. Pentosaccharide inhibits Xa factor more specifically.
This heparin test is a two-step anti-decatoactivity assay for accurate and sensitive determination of heparin concentrations in plasma or purified solutions. The plasma of the test sample needs to be diluted before analysis.
The purified human thrombin used in the experiment is predominantly present in the α form (obtained by direct activation of prothrombin) and exhibits higher coagulation reactivity than the "degraded" β or γ form under the condition of an equivalent concentration of chromogenic substrate.
The assay uses antithrombin and factor Xa reagents pre-diluted in specific buffers (not provided in this kit) and is compliant with the United States Pharmacopeia (USP) and the European Pharmacopoeia (EP).

Reagent composition


R1: Reagent 1 : Antithrombin III (human)
Antithrombin III (human), lyophilized with approximately 5 IU of activity per vial, containing BSA.
2 bottles (10mL bottle size).
R2: Reagent 2 : Activated Ten Factor (Bovine Source)
Activated 10-factor (bovine derived) lyophilized product containing approximately 40 μg (i.e., approximately 90 nkats, measured under optimized conditions for S-2765-specific substrate) activity per vial. Activated ten-factor concentrations are precisely adjusted for each lot to provide optimized assay reactivity and linearity.

R3: Reagent 3: Ten-factor-specific chromogenic substrate S-2765
Ten-factor-specific chromogenic substrate S-2765, each vial contains approximately 4 mg (approximately 6 μmol) of chromogenic substrate lyophilized product, containing mannitol.
